摘要
目的:观察脂多糖对人牙周膜成纤维细胞COX-2表达的影响,探讨牙周病发生发展过程中脂多糖(LPS)与COX-2的关系。方法:使用不同浓度LPS刺激体外培养人牙周膜成纤维细胞,免疫组织化学法检测细胞COX-2蛋白表达,灰度分析并进行统计学处理。结果:LPS组COX-2蛋白表达明显高于对照组,并且LPS浓度越高,COX-2表达越强。结论:LPS能够诱导人牙周膜成纤维细胞COX-2表达上调,随着LPS刺激浓度增高,COX-2表达逐渐增强,这一机制很可能在牙周炎发生发展过程中扮演重要角色。
Objective:To study lipopolysaccharide-induced cyclooxygenase-2 protein expression by the stimulate of lipopolysaccharide in human periodontal ligament fibroblasts(HPDLCs)and analyse the relationship with periodontitis.Method:HPDLCs was cultured in vitro,stimulated by lipopolysaccharide of varial viscosity,Then we test the expression of COX-2 by using immunohistoche staining and determination.Result:The expression of COX-2 with the stimulas of LPS is higher than the group of contrast.The thicker LPS is taken,the higher expression of COX-2 is.Conclusion:LPS may induce the expression of COX-2 in HPDLCs and with the thicker LSP taken,the higher expression of COX-2 is,which plays a very important role in periodontitis.
