摘要
目的建立高脂血症大鼠重症急性胰腺炎模型。方法SD大鼠脂肪乳剂灌胃2周建立高脂血症模型,逆行胰胆管注射3.5%牛磺胆酸钠诱发重症急性胰腺炎(SAP)模型。将雄性SD大鼠50只随机分为4组,即正常组(n=10)、HL组(n=10)、SAP组(n=15)、HAP组(n=15)。正常组均衡饲料饲养2周;HL组脂肪乳剂灌胃2周;SAP组逆行胰胆管注射3.5%牛磺胆酸钠溶液;HAP组脂肪乳剂灌胃2周+逆行胰胆管注射3.5%牛磺胆酸钠溶液。检测各组的血清淀粉酶、三酰甘油及胆固醇水平,观察胰腺组织病理改变。结果SAP组及HAP组血清淀粉酶活性较正常组明显升高(P<0.05),胰腺组织出现水肿、出血及坏死改变;HL组血清三酰甘油、胆固醇值较正常组明显升高(P<0.05),胰腺组织无明显的炎症改变;HAP组胰腺组织病理改变较SAP组严重。结论自制脂肪乳剂灌胃2周结合逆行胰胆管注射3.5%牛磺胆酸钠溶液可成功建立高脂血症大鼠重症急性胰腺炎模型;高脂血症可加重重症急性胰腺炎大鼠胰腺的病理改变。
Objective To establish a model of hyperlipemia rat with severe acute pancreatitis. Methods Rats received intragastric administration of high fat diet two weeks which induced experimental hyperlipemia, and retrograde injection of 3.5 % sodium taurocholate was injected into the pancreatic duct which induced severe acute pancreatitis. Fifty male SD rats were randomly divided into four groups: control group, HL group, SAP group, and HAP group. Control group were with balance feed for two weeks. HL group underwent intragastric administration with high fat diet for two weeks. SAP group underwent retrograde injection of 3.5% sodium taurocholate into the pancreatic duct. HAP group underwent intragastric administration with high fat diet for two weeks and retrograde injection of 3.5% sodium taurocholate into the pancreatic duct. The value of am- ylase (AMS), triglyceride (TG), and cholesterol (CH) in serum were measured, and the histopathologic damage degree of pancreas were observed. Results The serum AMS values in SAP group and HAP group were higher than that in control group (P〈0.05), oedema, hemorrhage and necrosis were detected under the light microscopy; the serum TG, CH in HL group were significantly higher than those in control group (P〈0.05), but no conspicuous inflammation present in pancreatic tissue; the histopathologic damage of pancreas in HAP group were more serious than those in SAP group. Conclusion A model of hy- perlipemia rat with severe acute pancreatitis might be developed by intragastric administration of high fat diet two weeks and ret- rograde injection of 3.5 % sodium taurocholate injected into the pancreatic duct. Hyperlipemia can aggravate the histopathologic damage of severe acute pancreatitis in rats
