摘要
论文采用硫酸铵分段盐析,DEAE-32纤维素柱层析等方法从东亚钳蝎蝎尾中提取出相对分子质量为37.7 k的单一蛋白,纤溶平板法证明其具有纤溶活性,并研究了温度、时间、pH值、金属离子对其溶纤活性的影响。实验结果表明蝎毒活性蛋白最适作用温度为35℃,最适pH值为5.0,Fe2+、Ca2+、Ba2+对其纤溶活性有促进作用,Cu2+、Mn2+对其有抑制作用,同时时间作用对其影响很小。并讨论其体外溶栓作用,表明其有溶栓活性。
A pure fibrinolyric protein SVP with 33. 7 k molecular weight, was isolated and purified from buthus martensii karsch by ammonium precipitation and DEAE ion exchange chromatography. Its fibrinolysis activity was proved by the fibrinolysis plate. The influences of temperature, time, pH value and metallic ions on its activity were studied. The results showed that its optimal temperature was 35 ℃, optimal pH value was 5. 0, and the enzyme activity was improved by Fe2+ and Ca2+ and inhibited by Ba2+ , Cu2+ and Mn2+. The results showed that time had little effect on its fibrinolysis activity, temperature was the most important factor. Its thrombolytic activity was also proved in this study and it has a strong thrombolytic activity on blood clot of chicken.
