摘要
目的:研究重组PAI-2(纤溶酶原激活剂抑制剂-2)基因在裸鼠皮下纤维肉瘤移植瘤细胞中的表达及其对超微结构的影响。方法:应用常规诊断电镜和免疫电镜技术,观察肿瘤细胞的结构变化,定位研究PAI-2蛋白的表达。结果:重组PAI-2基因转染的纤维肉瘤细胞,其移植瘤具有PAI-2蛋白的表达,以活性形式存在于细胞表面,同时细胞超微结构的恶性特征减弱,分化趋向成熟,侵袭转移能力下降。
Purpose To study the effect of recombinant PAI 2 on ultrastructure of nude mouse fibrosarcoma transplanted tumor. Methods The localization of PAI 2 protein and the ultrastructure of tumor cells were observed by routine diagnosis and immunoelectron microscopies. Results The fibrosarcoma transplanted tumor cells expressed PAI 2 protein which was located on the cell surface, whereas the ultrastructural malignant characteristics of tumor cells had abated with the tendency of well differentiation. Conclusion The expression of PAI 2 gene plays an important negative regulation role in the mechanism of tumor invasion and metastasis.
