摘要
目的:BHRF1的研究对于认识EBV有关肿瘤和其它疾病的发病机理、寻找预防、治疗途径有十分重要的意义。方法:以B958细胞DNA为模板,设计一对引物,用PCR方法扩增BHRF1基因,用目的基因内的两个限制性内切酶位点进行酶切鉴定。结果:扩增产物长592bp,与预期结果一致,两个酶所切下的四个片段长度也与理想值相符。结论:本研究首次采用PCR方法,成功地获得完整的EB病毒BHRF1基因,从而为该基因的进一步研究和应用奠定基础。
Objective:To study the function of BHRF 1 gene for elucidating the mechanism of EBV related tumours and other diseases,their prevention and treatment.Methods:The BHRF 1 gene was amplified from template DNA of B 95 8 cell by polymerase chain reaction,and was identified with method of digestion with two unique restriction endonucleases.Results:The amplified fragment was about 592 bp in length.The base pairs of these four bands which cleaved with two unique restriction endonucleases are in coincidence with lengthes as predicted.Conclusions:The intact BHRF 1 gene,for the first time,is obtained by using PCR technique and primers designed by ourselves.The study provides a model for further research on EBV BHRF 1 gene.
