摘要
目的:观察白细胞介素8(IL-8)在异种骨移植局部的表达及定位。方法:将牛松质骨载体植入小鼠股部肌肉内。于术后4、7、14和21天取材,应用原位杂交和免疫组织化学技术检测植骨局部IL-8mRNA及蛋白的表达与细胞定位。结果:术后4、7、14和21天,植骨局部均有IL-8mRNA及蛋白的表达。mRNA主要定位于载体周边的单核细胞和巨噬细胞,成纤维细胞、部分间充质细胞、血管内皮细胞也有阳性表达。免疫组织化学检测结果与原位杂交结果相一致。结论:在异种骨移植局部多种细胞表达IL-8mRNA及蛋白,提示IL-8在异种骨移植中与局部免疫反应密切相关。
bjective: To observe the expression and cellular localization of interleukin- 8 mRNA and protein in the area of xenogeneic bone implant. Methods: The bovine cancellous bone particles were implanted into the muscle of the legs of mice,and were removed 4,7,14 and 21 days later. IL-8 mRNA and protein in the site of implant were assayed by in situ hybridization and immunocytochemistry.Results:The expression of IL-8 mRNA and protein were observed in all specimens at 4,7,14 and 21 days after implantation.IL- 8 mRNA and protein express mainly by monocytes, macrophages, fibroblasts, and some of mesenchymal cells, vascular endothelial cells. Conclusion: Many different kinds of cells express IL-8 mRNA and secret IL-8 in the area of xenogeneic bone implant, it suggests that IL-8 may play an important role in local immunity of xenogeneic bone graft.
