RB基因在小鼠生殖细胞发生过程中的表达研究 被引量:3

STUDY ON THE EXPRESSION OF RB GENE DURING GAMETOGENESIS OF KUNMING MICE

摘要 为研究RB基因在小鼠生殖细胞发生过程中的表达,用地高辛(DIG)标记的RB基因的DNA探针和碱性磷酸酶标记的抗地高辛抗体,在组织切片上进行DNA-mRNA分子原位杂交分析方法,检测了昆明种正常雄性小鼠及雌性小鼠不同发育期生殖细胞发生过程中RB基因的转录表达(mRNA),结果发现:(1)在睾丸组织中,RB基因只在性未成熟组睾丸曲细精管的初级精母细胞,精子细胞层以及性成熟组睾丸曲细精管的精子细胞层中有强转录表达;在精原细胞和生精干细胞层以及成熟精子细胞中都不表达;(2)成熟卵巢中,RB基因在次级卵泡的卵母细胞中,以及孕期卵巢的次级卵泡的卵母细胞核中均有强度不同的转录表达;初级卵泡的卵母细胞中没有表达,成熟卵泡中的卵母细胞中也不再有表达。这些结果表明。 Using in site hybridization on tissue section with Dig DNA labeling and detection system, We studied the expression of the antioncogene RB in testis and ovary of Kunming mice. The results are as follows:(1)in testis tissue, the expression of RB gene only was found in spermatocytes and prespermatids of seminiferous tubule of immature mice and spermatids of mature mice, and it was not found in the spermatogonia, spermatognial stem cell and primary metamorphous sperm from both immature and mature mice. (2)In ovary tissue, the expression of RB gene was found in oocytes of second follicle, the strength of expression was of variations with different phases of development, and it was not found in both primary oocytes and mature oocyte. The expression of RB gene was also found in oocyte nucleus of second follicle in pregnant mice. These results showed that the RB gene might involve in the spermatogenesis and oogenesis of mice.
