96孔板高通量筛选多杀菌素高产菌株的研究 被引量:16

摘要 考察了刺糖多孢菌在96孔板固体培养基和液体培养基中的生长情况,建立了96孔板高通量筛选多杀菌素高产菌株的方法。结果表明:96孔板三明治盖能同时满足过滤杂菌和刺糖多孢菌生长耗氧所需,刺糖多孢菌在96孔板固体培养基中的生长情况与传统斜面培养相当,种子培养最适种龄为60h,每孔发酵培养基装液量为0.4mL。与传统摇瓶发酵筛选平台相比,该方法能显著提高多杀菌素高产菌株的筛选效率。 Based on the growth of Saccharopolyspora spinosa in both solid and liquid medium,a high throughput method was established to screen high spinosad-producing strains with 96-well plates.The results showed that applying the sandwich covers for 96-well plates could meet the requirement of bacteria filtration and oxygen supply for the growth of S.spinosa.S.spinosa grew well in both 96-well plate and slant culture.The optimum seed growing time was 60 h;the broth content of each well was 0.4 mL.Comparing with the traditional shake flask screening platform,this method could significantly improve the screening efficiency of high spinosad-producing strains.
