摘要
目的研究胃癌细胞中WNT&HH信号通路的内在性关系。方法用RT-PCR方法分别检测不同浓度(50μmol/L,75μmol/L,100μmol/L)环靶明干预的胃癌细胞(SGC-7901)24 h后的β-can-tenin mRNA、Gli1 mRNA、Sfrp1 mRNA的表达。结果 1、胃癌细胞梭型贴壁生长,染色质分布均匀,细胞膜完整;2、环靶明干预胃癌细胞后,胃癌细胞的生长明显抑制。Gli1 mRNA的表达呈抑制浓度依赖型,Sfrp1 mRNA的表达随Gli1 mRNA的表达减少而加强,β-cantenin mRNA的表达随Sfrp1 mRNA的表达增加而减少。结论 Gli1 mRNA和Sfrp1 mRNA之间有关联性。
Objective To investigate the interrelation between HH and WNT signal pathway. Methods To examine the expression of β-cantenin mRNA Glil mRNA Sfrpl mRNA in Gastric Caner Cell with Cyclo- pamine blockage in 24 h respectively. Results With Cyclopamine ' s intervention, the number of Gastric Canc- er Cells has been restrained. The expression of Glil mRNA has fallen with the increase of concentration; the ex- pression of Sfrpl mRNA has increased with the decrease of the expression of Gliland the expression of β-cante- nin mRNA has decreased with the increase of the expression of Sfrpl mRNA. Conclusion Glil mRNA Sfrpl mRNA would be interrelated.
