奶牛NUCB2基因突变及其与产奶量关联的分析

摘要 利用PCR-SSCP和DNA测序技术首次研究了中国荷斯坦牛NUCB2基因的变异。结果表明在NUCB2基因外显子9发现两个沉默突变:27451G>A和27472T>C,两个碱基变化没有导致氨基酸的改变,但两个位点沉默突变同时发生,表现出3个基因型,即MM、MN和NN,经过对不同基因型与奶牛产奶量性状的关联分析,发现这些突变对奶牛产奶量产生显著影响(P<0.05)。 The mutations of NUCB2 gene were studied with PCR-SSCP and DNA sequencing methods in Chinese Holstein. The results showed that two mutant sites with g27451G〉A and g27472T〉C were detected only at the exon9 of NUCB2 gene. These two silent mutation did not result in the change of amino acid, but they happened simutaneously. They showed three genotypes, MM, MN and NN. The different genotypes of these two mutation loci were significantly associated with milk production (P〈0.05).
