摘要
目的通过免疫组化、原位杂交等实验方法观察前癃通对大鼠前列腺增生组织中内皮生长因子(VEGF)表达的影响,从转录和翻译水平探讨其治疗的分子机制,促进前癃通治疗良性前列腺增生症中推广应用。方法60例SD大鼠分成空白对照组,模型组和前癃通低、中、高剂量组和癃闭舒组,采用免疫组化和原位杂交的方法观察各组大鼠前列腺增生组织中VEGF阳性表达情况(所有图像均采用MIAS图像分析系统观察分析)。结果6组大鼠前列腺组织中VEGF平均灰度值、积分光密度、阳性细胞总面积比较,差异均有统计学意义(P<0.01)。模型组显著高于空白对照组(P<0.01),前癃通低、中、高剂量组及癃闭舒组大鼠前列腺组织中VEGF各检测指标与模型组比较,差异均有统计学意义(P<0.01),前癃通高剂量组灰度值、积分光密度、阳性细胞总面积均低于癃闭舒组,差异有统计学意义(P<0.05)。结论前癃通可以降低大鼠前列腺组织中VEGF的阳性表达,高剂量较低剂量的影响更明显。
Objective To explore the effect of traditional Chinese drug Qian Long Tong on the expression of vascular endothelial growth factor （VEGF） in rats with benign prostatic hyperplasia.Methods Totally 60 SD rats were equally randomized into blank control group,model group,low-,intermediate-and high-dose Qian Long Tong groups,and Longbishu （another traditional Chinese drug with similar efficacy） group.The expression of VEGF in the benign prostatic hyperplasia was detected using immunohistochemical method and in situ hybridization.All the pictures were analyzed with the MIAS images analysis system.Results The mean gray level,integral optical density,and positive cell area of VEGF in the prostate tissues were significantly different among these 6 groups （P〈0.01）.They were significantly higher in model group than in blank group（P〈0.01）.VEGF-related indicators in low-,intermediate-and high-dose Qian Long Tong groups and Longbishu group were significantly different when compared with the model group （P〈0.01）.The mean gray level,integral optical density,and positive cell area of VEGF were significantly higher in high-dose Qian Long Tong group than in Longbishu group （P〈0.05）.Conclusion The traditional Chinese drug Qian Long Tong capsule can decrease the expression of VEGF in benign prostatic hyperplasia and improve the pathologic changes of the prostate tissues.
