实时定量PCR检测蛋壳形成时骨桥蛋白表达变化的研究 被引量:1

摘要 骨桥蛋白(osteopontin,OPN)是一种具有多种功能分泌型钙结合磷酸化糖蛋白。为了从基因表达的角度来解析蛋鸡蛋壳形成的分子机理,本研究以产蛋鸡为试验动物,采用实时定量PCR技术对与蛋壳形成有关的骨桥蛋白基因在子宫中不同时间段(蛋进入子宫前2 h、进入子宫前1 h、进入子宫后5 h、进入子宫后15 h、产蛋后1 h、产蛋后2 h)的表达丰度进行了研究。结果表明,在蛋进入子宫前2 h、蛋进入子宫前1 h、蛋进入子宫后5 h OPN的表达量逐渐升高,之后在蛋进入子宫后15 h、产蛋后1 h、产蛋后2 h又逐渐下降,这一规律性的变化与蛋壳的钙化密切相关。 Osteopontin(OPN) is a kind of multi-functional secretory calcium-binding phosphorylated glycoprotein.In order to resolve the molecular mechanism of the formation of eggshell from the perspective of gene expression,egg-laying hens as the experimental animals in this study,using real-time quantitative PCR technique,to study the abundance expression of the osteopontin gene which was related to eggshell formation in the uterus at different time periods(2 h before egg into the uterus,1 h before into the uterus,5 h after into the uterus,15 h after into the uterus,1 h after egg production,2 h after egg production).The results showed that 2 h before egg into the uterus,1 h before into the uterus,5 h after into the uterus,OPN expression levels increased gradually;15 h after into the uterus,1 h after egg production,2 h after egg production OPN expression levels decreased gradually.The change of regularity was closely related to egg shell calcification.
