摘要
目的观察高压氧治疗对脊髓损伤后SD大鼠的神经细胞凋亡及行为学评分的影响。方法雄性SD大鼠48只,随机分为高压氧治疗组、对照组及伪手术组,每组各16只大鼠(n=16),每组又分为4个时间点,分别为脊髓损伤后6、12、24 h和72h,其中每个时间点高压氧治疗组大鼠4只,对照鼠4只和伪手术鼠4只。高压氧组和对照组采用脊髓横切法造成脊髓损伤模型,高压氧治疗组大鼠在每个时间点给予高压氧治疗后即处死,对照组和伪手术组在伤后相应时间点处死。用脱氧核糖核苷酸末端转移酶介导的缺口末端标记法标记凋亡细胞,用改良的TARLOV评分评估大鼠后肢运动功能。观察比较各时间点的凋亡指数和改良的TARLOV评分的差异。结果伪手术组未见凋亡阳性细胞;凋亡细胞在伤后6 h即出现在脊髓组织中,24 h与72 h凋亡细胞数显著增加;与对照组相比,高压氧治疗组的TUNEL阳性细胞显著减少(P<0.05);同时,高压氧组大鼠改良的TAR-LOV评分显著高于对照组(P<0.05)。结论高压氧治疗能够显著减少脊髓损伤后大鼠神经细胞凋亡并改善大鼠后肢运动功能,对脊髓损伤后的神经细胞具有很好的保护作用。
Objective To investigate effects on hyperbaric oxygen(HBO) on apoptosis of nerve cells and motor function after spinal cord injury. Methods 48 SD male rats were randomly divided into three groups: hyperbaric oxygen group (n=16) ,control (n=16)and sharn operate group (n=16). Rats sacrified at 6 hours,12hours,24 hours and 72 hours post injury,among which rats in HBO group were received HBO therapy before killing at each lime point. Apoptosis index and improved Tarlov's score were calculated at caeh time point. Results No apoptotic cell appeared in sham group. The apoptosis cell could be observed at 6 hours post-injury,and elevated from 24 to 72 hours. Compared to HBO group, the TUNEL-positive cells were significantly higher in the control group(P〈0.05). Improved Tarlov's score was lower in control group than that of HBO group (P〈0.05). Conclusion HBO therapy can reduce and protect nerve cell from apoptosis after spinal cord injury in rats and improve their motor function.
