摘要
目的研究趋化因子受体CXCR4的mRNA及其蛋白在喉鳞状细胞癌(简称喉癌)的表达,分析该表达与喉癌临床病理特征的关系,探讨CXCR4在喉癌发生、发展、转移中的作用及临床意义。方法应用逆转录聚合酶链反应及免疫组织化学技术,测定42例新鲜喉癌黏膜及癌旁黏膜的CXCR4的mRNA和蛋白的表达,并与患者临床病理特征相比较;应用Image-pro-plus软件对肿瘤细胞光密度定量分析。结果CXCR4在喉癌中呈过度表达,喉癌旁黏膜弱表达或不表达,两种黏膜差异有显著性意义(P<0.01);CXCR4的表达与患者的性别、年龄、病变部位、T分级无关(P>0.05);与病理分级相关(P<0.01),与是否有淋巴结转移相关(P<0.01)。结论CXCR4在喉癌黏膜中的mRNA及蛋白表达率显著高于正常黏膜,且与肿瘤的病理分级和淋巴结转移相关,对喉癌的发生、发展和转移起重要作用。
OBJECTIVE To investigate the mRNA expression of CXCR4 in laryngeal squamous cell carcinoma and its correlation with clinicopathologic parameters, and to evaluate its role in the carcinogenesis and progression of laryngeal squamous cell carcinoma. METHODS The mRNA expression of CXCR4 in fresh laryngeal squamous cell carcinoma tissues and adjacent normal tissues from 42 patients were examined by reverse transcription-polymerase chain reaction(RT-PCR).The protein expression of CXCR4 were detected by immunohistochemical staining method and analyzed with Image-pro-plus software. RESULTS The positive expression rate of CXCR4 in tumor samples were significantly higher than that in normal tissues(P0.01). In tumor samples, the expression was not associated with age, sex, tumor site and T stage(P0.05). The expression was higher in tumors of stage Ⅲ, IV than that in stage I, Ⅱ(P0.01). The expression was significantly higher in tumors with cervical lymph node metastasis than that without cervical lymph node metastasis(P0.01). The expression of CXCR4 protein and CXCR4 mRNA had a positive relationship. CONCLUSION The expression of CXCR4 may play an important role in the development and lymph node metastasis of laryngeal squamous cell carcinoma. CXCR4 produced by tumor cells may up-regulate the microvascularization in the laryngeal squamous cell carcinoma, and enhance the growth, spreading and lymph node metastasis in human laryngeal squamous cell carcinoma. Its expression in laryngeal squamous carcinoma may help us to judge the malignant degree and metastasis potential of the tumor.
