摘要
以人参地土壤为材料,比较研究SDS高盐加变性剂法、SDS高盐法和试剂盒法等3种土壤微生物基因组DNA提取方法.结果表明,3种方法均可提取人参地土壤微生物基因组DNA,但每种方法所获得的DNA在颜色、提取量及纯度等方面都存在较大差异.普通手工方法(SDS)提取的DNA由于纯度不高等原因很难进行后续的PCR扩增,试剂盒法能够在较短时间内获得用于PCR扩增的较高纯度的DNA,所以该方法是较好的土壤微生物DNA提取方法.
Compaired three microbial DNA extraction methods of Ginseng soil. The results showed that all of the three methods can extract Ginseng soil microbial DNA, but the color, yield, purity and the effect of PCR amplification differ within the three methods. Comparatively, although the DNA yield of the third method (using DNA extraction kit)less than the former, but the DNA more suitable in PCR reaction.
