摘要
采用改进的CTAB法提取朝鲜白头翁的基因组总DNA,建立朝鲜白头翁RAPD分析的PCR反应体系,成功进行RAPD扩增.筛选出的最佳PCR反应体系为:25μL反应体系中包括模板DNA 20ng,dNTP 200μmol/L,引物0.4μmol/L,Mg2+1.5 mmol/L,Taq酶1 U,10×Buffer 2.5μL,其余部分为无菌重蒸馏水.
The genomic DNA from the young of Pulsatilla cernua (Thumb.) Bercht. was successfully extracted by using the developed CTAB method and a steady system of PCR reaction for P. cernua was established. The optimal system 20 ng DNA,200μmol/L dNTP,0.4μmol/L random primer, 1.5 mmol/L Mg2+ ,1 U Taq polymerase,2.5 μL 10×Buffer in 25 μL reaction solution.
