摘要
以80%的冷甲醇浸提烟叶样品中的生长素(IAA)、脱落酸(ABA),通过控制pH值,经过多次液液分配并结合聚乙烯吡咯烷酮(PVP)和C18筒柱进一步分离纯化,用反相液相色谱检测烟叶成熟过程中内源IAA、ABA的动态变化。结果表明:下部叶叶色黄绿,黄色明显,主脉1/2~2/3白,支脉1/3白;中部叶叶色黄绿以黄为主至淡黄,主脉近全白,支脉近2/3白等外观特征出现时,叶片进入迅速衰老时期,超过上述时期的叶片应为过熟。
IAA and ABA in tobacco leaf samples were diffused with 80% cool methanol.By controlling pH value,after several times of liquid liquid partition and further separation and purification with PVP and C 18 column,changes of IAA and ABA during maturing were tested by reversed phase HPLC.The results suggest that tobacco leaves enter quick senescence period when the following external property appears:greenish yellow lower leaves are notably yellow with 1/2 ̄2/3 of midrib and 1/3 of lateral vein in white,greenish yellow middle leaves turn mainly yellow to light yellow,with midrib nearly all in white and 2/3 of lateral vein in white.The leaves exceeding that period are over mature.
