摘要
本文设计了大鼠β-肌动蛋白基因的一对 引物,用冰冻组织切片RT-PCR方法,检测了鼠肝、肾该基因的mRNA,结果能够扩增该基因的cDNA片段和基因组片段。用这种新的方法进行RT-PCR检测组织的mRNA,快速、简单,同时也能够避免组织RNA的降解、污染。该方法也能直接PCR检测基因组的DNA片段。
Using a specific primers for rat β-actin gene. we have detected rat liver and kindey tissue β-actin gene mRNA by RT-PCR method using cryosections. Both the cDNA and genomic DNA of the gene was detected. As a new method to detect tissue mRNA, it is rapid, simple and can avoid RNA degrading. In addition, using this method we can directly amplificate genomic DNA.
