摘要
目的:观察风湿性心脏病人瓣膜成纤维细胞的胶原合成,为研究风湿性心脏病瓣膜纤维化的发病机制建立基础。方法:采用消化组织块培养法建立人二尖瓣和主脉瓣成纤维细胞体外培养方法,对5例风湿性心脏病人和5例尸检中除外心血管疾病和胶原系统疾病的二尖瓣和主动脉瓣成纤维细胞进行了体外培养,并用3H-脯氨酸测定瓣膜成纤维细胞的胶原合成。结果:二尖瓣和主动脉瓣的瓣膜成纤维细胞其形态、生长方式及胶原合成差异均不明显,风湿性心脏病瓣膜成纤维细胞的胶原合成明显高于对照组。
Objective: To observe the collagen synthesis of cultured cardiac valve fibroblast from patients with rheumatic heart disease to lay the foundation for further studies on the mechanism of fibrosis of the rheumatic cardiac valves. Methods: Using enzyme digestion, the mitral and aortic valve fibroblasts from 5 patients with rheumatic heart disease and other 5 confirmed by autopsy that they did not die of cardiovascular disease and collagen system disease were cultured in vitro . The collagen synthesis was determined with 3H proline incorporation. Results: There was no difference in cell shape, proliferation and collagen synthesis between the mitral valve fibroblast and aortic valve fibroblast. Collagen synthesis of the rheumatic valvular fibroblasts was significantly higher in 5 patients with rheumatic heart disease than in the other 5. Conclusion: Collagen synthesis of the rheumatic cardiac valvular fibroblasts cultured in vitro is increased.
