摘要
本研究采用四甲基偶氮唑(MTT)比色法研究了AFB1和AMN在不同添加剂量和暴露时间下对Caco-2细胞增殖的影响,同时对Caco-2细胞用作研究AFB1转运和吸收模型的可行性进行初步评估。结果表明:在AFB1添加剂量为250、500、750、1000μg/L和暴露时间为12、24、36、48h的情况下,与阴性对照组(培养基)相比,AFB1均显著降低了Caco-2细胞的相对存活率(P<0.05),且呈一定的剂量和时间效应关系。在AMN添加剂量为0.25%、0.5%、0.75%、1.0%和暴露时间为24、48、72h的情况下,与阴性对照组(培养基)相比,AMN材料本身虽然也显著降低了Caco-2细胞的相对存活率(P<0.05),但Caco-2细胞的相对存活率均在88.76%以上,毒性级别仅为1级。在AMN不同添加剂量和暴露时间为48h的情况下,与阳性对照组(900μg/LAFB1)相比,AMN均显著提高了Caco-2细胞的相对存活率(P<0.05)。结果提示,AMN能有效吸附AFB1并显著降低其对Caco-2细胞的毒害;AFB1对Caco-2细胞具有显著的增殖毒性,Caco-2细胞不宜直接用作研究AFB1转运和吸收的体外模型。
This experiment was conducted to study the effect of Al2O3-pillared montmorillonite nanocomposites(AMN) on the protection of Caco-2 cells against cytotoxicity induced by AFB1 and whether Caco-2 cells model could be engaged in AFB1 absorption research.MTT method were performed to investigate the effect of AFB1 and AMN on Caco-2 cell proliferation.At levels of 250,500,750,1 000 μg/L of AFB1 and duration of exposure of 12,24,36,48 h,relative survival index of Caco-2 were greatly decreased(P〈0.05).Dose-effect relationship was observed.Addition of AMN at levels of 0.25%,0.5%,0.75%,1.0% and duration of exposure of 24,36,48 h,the relative survival index of Caco-2 maintained all above 88.76%.The cytotoxicity level of AMN on Caco-2 was at Calss I,meeting the conditions required by the law of the People′ s Republic of China national standards on human implantation materials.Compared with the positive control of(900 μg/L AFB1),addition of AMN at levels of 0.25%,0.5%,0.75%,1.0% and duration of exposure of 48 h,significantly increased the relative survival index of Caco-2 cells(P〈0.05).The results indicated that AMN could prominently decrease the damage caused by AFB1 on Caco-2 cells,while Caco-2 cells model was not supposed to be directly used in AFB1 absorption study.
