摘要
目的探讨GRP78与HSP60蛋白在原发性多结节肝癌组织中不同克隆来源的表达,以及与原发性多结节肝癌侵袭、转移的关系。方法采用免疫印迹法检测10例单中心性、10例多中心性、7例单结节、5例正常肝组织中GRP78与HSP60蛋白表达。结果GRP78与HSP60蛋白的相对表达量在单中心性组[(0.42±0.02)INT/mm2、(0.39±0.04)INT/mm2]、多中心性组[(0.34±0.01)INT/mm2、(0.32±0.05)INT/mm2]、单结节组[(0.32±0.04)INT/mm2、(0.31±0.03)INT/mm2]、正常组[(0.20±0.04)INT/mm2、(0.20±0.04)INT/mm2]依次减少(P<0.05)。结论GRP78和HSP60在多结节肝癌不同克隆来源的组织中表达有差异,过度表达的HSP蛋白可能与多结节肝癌的发生有关,对于鉴定多结节肝癌克隆来源有一定的参考价值。
Objective To investigate the expression of glucose-regulated protein(GRP)78 and heat shock prteins(HSPs)60 in multifocal hepatocellular carcinoma,as well as their clinical significance.Methods Western blot was used to detect the expression of HSPs in 10 cases of intrahepatic metastasis(IM group),10 cases of multicentric occurrence(MO group),7 cases of single nodule(S group)and 5 cases of normal tissues(N group).Results According to the results of western blot,HSPs expression level were reduced in the order from IM group[(0.42±0.02 INT/mm^2,(0.39±0.04) INT/mm^2]to MO group[(0.34±0.01)INT/mm^2,(0.32±0.05)INT/mm^2],S group[(0.32±0.04)INT/mm^2,(0.31±0.03)INT/mm^2]and N group[(0.20±0.04)INT/mm^2,(0.20±0.04)INT/mm^2](P0.05).Conclusion Expression of GRP78 and HSPs are different in different clone-drive tissue of multifocal hepatocellular carcinoma,Overexpression of may play a role in the genesis of multifocal hepatocellular carcinoma and in indentifying its clone-drive.
