一贯煎对肝炎小鼠模型TNF-α信号通路的影响 被引量:4

摘要 目的:探讨中药一贯煎对肿瘤坏死因子α(TNF-α)致肝炎小鼠模型TNF-α信号通路的干预作用。方法:将C57BL/6小鼠随机分成4组,正常组、模型组、中药组、阳性对照组。正常组及模型组小鼠灌服生理盐水。中药组给予一贯煎汤药灌胃。阳性对照组给予联苯双酯滴丸灌胃。均每天1次,持续1周。模型组、中药组、阳性对照组小鼠腹腔注射0.1μgTNF/12mgD-氨基半乳糖(GaIN)制造肝炎小鼠模型,进行肝功能、肝组织病理、WesternBlot检测TNF-α信号通路相关蛋白cIAP1。结果:一贯煎能减轻肝细胞炎症反应、抑制肝细胞的凋亡、坏死,使小鼠体内ALT和AST水平显著下降(P<0.05);能促进肝细胞cIAP1蛋白的表达(P<0.05)。结论:一贯煎治疗肝炎的药理机制之一在于调节TNF-α信号通路相关蛋白的表达,从而有效减轻炎症反应,达到保肝、降酶的目的。 Objective: To investigate the effects of Yiguanjian on TNF-α signal transduction in hepatitis mouse induced by TNF-α. Methods: C57BL/6 mouses were randomized to normal control group, model group, TCM group and positive control group. Normal control group and model group were intragastrically with saline, once a day for a week. TCM group were intragastrically with Yiguanjian, once a day for a week. Positive control group were intragastrically with Bifendate Pills, once a day for a week. Model group, TCM group and positive control group were intraperitoneally injected with 0.1μg TNF/12mgGaIN per mouse to induce hepatitis. All mouses were killed when one mouse began to appear amord situation. Blood samples were taken for the detection of liver function. Liver samples were taken for the observation of pathology and the test of cIAP1 protein. Results: Yiguanjian could lessen the degree of liver inflammation, restrain the apoptosis and necrosis of liver cell, ALT and AST obviously decreased (P0.05); cIAP1 significantly increased (P0.05). Conclusion: One of mechanisms of Yiguanjian which prevent liver damage and lower the serum transaminase level was to adjust the expression of some protein of TNF-α signal transduction.
