摘要 目的研究下丘脑和海马内c-fos和c-jun蛋白的表达与应激反应之间的时效关系。方法采用特异性抗体的原位免疫细胞化学方法,对游泳应激后不同时间点的大鼠下丘脑和海马内fos和jun蛋白阳性神经元的分布进行观察,用图像分析技术对大鼠下丘脑室旁核(PVN)、视上核(SON)和海马齿状回(DG)内的fos和jun阳性细胞的相对切面面积比和平均目标灰度进行分析。结果游泳应激结束后3h,室旁核和视上校及齿状区内可见密集深染的c-fos和c-jun阳性细胞,室旁核小细胞部,背内侧核等区域亦有疏密不一的阳性胞体。图像分析结果发现,游泳应激结束后3~8h内下丘脑PVN和SON及海马DG内fos和jun阳性胞体的相对切面面积比明显高于正常大鼠(P<0.01),而平均目标灰度则显著降低(P<0.01)。结论应激可以诱导下丘脑和海马等相关脑区c-fos和c-jun的表达,并且具有很强的时效关系。 Objective To explore the possible time-effect relationship between stress response and expression of c-fos and c-jun in hypothalamus and hippocampus. Methods The immunohistochemistry was performed to observe the distribution of c-fos and c-jun netirons within hypothalamus and hippocampus, and the computerized image techniqlle was used to analyze the change in grey degree and relative sectional area of fos and inn-positive neurons in the hypothalamic paraventricular nucleus (PVN), the supraoptic nucleus (SON) and hippocampal dentate gyrus. Results A dense accumulation of fos and inn-positive neurons exhibited in the PVN, SON and DG, and some of fos and jun-positive neurons were also present in otherhypothalamus and hippocampus areas including parvocellular PVN (pPVN), dorsornedial nucleus and lateral hypothalamic area (LH). With using computerized image analysis, the relative sectional area of fos and inn--positive neurons in PVN, SON and DG at 3 ~8hafter stress were larger than that of normal group (P <0. of ), and the grey degree of fos and inn-positive neurons in PVN, SON and DG at 3~8h after stress were significant lower than that of normal group (P <0. of). Conclusion it is suggested that stress can induce the expression of c-fos and c-jun, and there are time--effect relationship between the stress response and expression of c-fos and c-jun.
