随机扩增多态DNA技术用于大肠杆菌基因分型的研究 被引量:1

A STUDY ON THE GENOTYPE OF ESCHERICHIA COLI ISOLATES BY RANDOM AMPLIFIED POLYMORPHIC DNA.

摘要 采用随机扩增多态 DNA(Random amplified polymophic DNA,简称 RAPD)技术,用二组引物对8株大肠杆菌进行了基因分型,发现二组引物均能将8株菌分成4个基因型,其中血清型相同,但来自不同国家和地区的3株被分成二个基因型;表明这三株菌基因组 DNA 间存在差异。4株从同一病人体内不同次分离到的血清型相同的菌株呈同一指纹图谱,说明该4株菌渊源于同一菌株。结果显示该方法的可分型性、敏感性、重复性及特异性均优于传统的微生物表型分型方法,是一种快速、简便的微生物基因分型方法,可广泛用于流行病学研究。 Random amplified polymorphie DNA(RAPD)bacterial fingerprinting was used for the genotype of es- cherichia coli with two primers.The results showed that the method was able to correctly distinguish the 2 of 3 epidemiological unrelated strains with the same serotype,4 strains from the same patient with the same serogroup yielded similar banding patterns and the strain with a different serotype gave unique fingerprint.The typability, sensitivity,reproducibility and specificity of PAPD is much better than that of traditional phenotypic techniques. RAPD is a rapid and simple genotypic technique.
