荧光定量RT-PCR检测类风湿关节炎患者葡萄糖-6-磷酸异构酶mRNA表达水平的研究 被引量:2

摘要 目的通过比较分析葡萄糖-6-磷酸异构酶(G6PI)mRNA在类风湿关节炎(RA)、其他自身免疫性疾病及健康人群之间的表达差异,探讨其在RA中的致病机制。方法建立实时定量RT-PCR方法,并利用该法检测128例RA患者(RA组)、32例其他自身免疫性疾病患者(疾病对照组)、100例健康体检者(健康对照组)G6PI基因的表达水平。结果经GAPDH校正后G6PI基因表达水平在RA组为2.72±2.09,其中活动组和缓解组分别为3.12±2.40和2.13±1.34,疾病对照组为2.14±1.43,健康对照组为1.25±0.73。RA组显著高于健康对照组(P<0.01),RA活动组基因表达水平显著高于缓解组(P<0.01),也显著高于疾病对照组(P<0.01)。结论 G6PI在RA患者中表达的显著增加表明G6PI可能参与了RA的发病过程,并与疾病的活动性相关,可作为RA的辅助性诊断指标及早期筛查指标。 Objective To invesligate the expression of mRNA of the glucose-6-phosphate isomerase(G6PI) in rheumatoid arthritis(RA) ,non-RA patients and healthy controls to preliminarily study the pathogenic mechanism of RA. Methods A real-time quantitative reverse transcriptase polymerase chain reaction(RQ RT-PCR) method was established for detecting G6PI and internal reference GAPDH expression levels in 128 RA patients,32 non RA patients and 100 healthy controls. Results The level of G6PI mRNA in RA patients 2.72±2.09 was higher than that of the healthy controls(1.25±0.73)(P〈0.01). In RA patients,the expression level of G6PI mRNA in the active stage(3.12±2.40) was higher than that at the stable stage(2. 13±1.34) (P〈0.01). Conclusion The increased expression of G6PI may play a pathological role in the development of RA,and it may be correlated with the activity of RA. G6PI, which can be regarded as a marker for early detection of RA.
