摘要
脂多糖(Lipopolysaccharide,LPS)能够通过上调转移相关因子白介素8(Interleukin-8,IL-8)基因的表达促进癌细胞转移.为探究LPS对转移能力不同人卵巢癌细胞IL-8mRNA表达的影响,我们以人低转移卵巢癌细胞株HO-8910和人高转移卵巢癌细胞株HO-8910PM为实验材料,采用实时定量反转录-聚合酶链式反应技术(qRT—PCR)研究LPS刺激后,IL-8 mRNA表达的变化.实验结果表明:与HO-8910相比,在HO-8910PM中LPS显著上调IL-8基因的表达水平(P〈0.05).我们推测LPS刺激IL-8基因在转移表型不同的卵巢癌细胞中表达的差异与癌细胞转移能力相关.
Up - regulation of metastatic - related gene of IL - 8 induced by LPS contributes to metastatic progression in various types of cancer. To investigate the relationship between LPS and production of IL - 8 mRNA in ovarian caner cells, the aherative expression of IL -8 induced by LPS was examined by quantitative reverse transcription - polymerase chain reaction ( qRT - PCR) in highly and lowly metastatie human ovarian cancer cell lines HO- 8910PM and HO -8910. We found that LPS significantly promoted production of IL- 8 mRNA in HO -8910PM cell line in contrast to HO-8910 cell line (P 〈 0.05 ). The results indicate that modification of IL- 8 mRNA level by LPS may has a relevance to metastatic ablilities of ovarian cancer cells.
