摘要
目的:建立聚乙二醇-2000(PEG)-(NH4)2SO4双水相体系萃取光度法测定亚甲基蓝(MB)的新方法。方法:测定了PEG相中亚甲基蓝吸收光谱和荧光光谱;研究了溶液酸度、盐用量、PEG用量及共存物质对体系测定的影响。结果:在溶液酸度pH7、PEG用量5.0 ml、硫酸铵加入量2.0 g、测定波长669 nm时,PEG相中MB有最大吸光度。线性回归方程Y=0.01929+0.11261X,相关系数r=0.9995,线性范围为0.08-1.50μg/ml。结论:方法具有快速、灵敏、简便的优点,用于天然水和针剂亚甲基蓝测定取得较好结果。
Objective:To establish a new method for spectrophotometric determination of methylene blue in polyethylene glycol-2000-(NH4)2SO4 aqueous two-phase system.Methods:The absorption spectrum and fluorescence spectrum of MB in PEG phase was detected.The effects of solution acidity,the dosages of PEG,salt and interference of other coexistence substances on the extraction determination of MB were studied.Results:In the polyethylene glycol phase of pH7,the 5.0 ml PEG and 2.0 g(NH4)2SO4,the MB had high absorbance with the maximum absorption of 669 nm.Beer′s law was obeyed in the range of 0.08-1.50 μg/ml for MB with the standard curve of Y=0.01929+0.11261X and correlation coefficient of 0.9995.Conclusion:The method is rapid,sensitive and simple.It has been applied to the determination of methylene blue in natural water and injection with satisfactory results.
