摘要
目的:对比观察老龄大鼠和成年大鼠转化生长因子-β1(TGF-β1)、肿瘤坏死因子-α(TNF-α)在博来霉素诱导肺纤维化模型中表达水平的变化,探索老年肺纤维化发病率升高的可能机制。方法:大鼠气管内灌注博来霉素(模型组)或生理盐水(对照组),于第7、14、21、28天分别处死5只,测定血清中TGF-β1、TNF-α的浓度;测定肺组织匀浆中羟脯氨酸(HYP)的浓度;通过HE染色观察肺组织形态的变化。结果:各时间点模型组大鼠肺组织HYP含量、血清中TGF-β1、TNF-α的浓度均较对照组明显增高,差异有显著性(P<0.05);成年和老龄模型组第7天至第28天肺组织HYP含量、血清中TGF-β1、TNF-α水平逐渐升高,但是成年模型组第21、28天与同组第14天比较差异无显著性(P>0.05),而老龄模型组在第21天和第28天时仍明显继续升高,且与同组第14天比较差异有显著性(P<0.05)。结论:气管内灌注博来霉素后老龄大鼠HYP、TGF-β1、TNF-α的浓度长时间及高水平表达,是肺纤维化随年龄增加的分子基础,也是老年人肺纤维化发病率升高原因之一。
Objective The difference of TGF-β1,TNF-α expression levels between adult rats and aged rats were compared to investigate the underlying mechanisms of the higher pulmonary fibrosis morbidity rate in the aged population. Methods The rats were either treated with a single intratracheal bleomycin injection (model group) or a 0.9% NaC1 injection (control group). 5 animals were killed on the 7th,14th,21st,28th days separately. Determined concentrations of TGF-β1 and TNF-α in serum and hydroxyproline (HYP) in lung tissue of rats. Observed morphological changes in lung tissue through the HE staining. Results The levels of HYP in lung tissue of rats and concentration of TGF-β1 and TNF-α in serum was significantly higher in the model group than those in the control group (P〉0.05); These concentrations increased gradually from the 7th to 28th in both adult rats and aged rats of the model group ,but there was no significant difference (P〈0.05) between the 14th day and the 21st,28th day in adult rats of model group,contrarily there was such significant difference in aged rats of model group (P〈0.05). Conclusion The prolongation and up regulation of HYP,TGF-β1 and TNF-α concentrations might contribute to the higher morbidity rate in the aged population and pulmonary fibrosis in the elderly.
