摘要
通过对9个牡丹品种叶片、叶柄总酚含量及多酚氧化酶活性的变化的研究,探讨了叶片、叶柄的总酚物质的含量和多酚氧化酶的活性与褐变的关系。结果表明:9个品种的叶片、叶柄总酚含量相差很大,叶柄的总酚含量比叶片低;叶片中总酚含量的顺序:似荷莲>萍实艳>洛阳红>丹皂镏金>金星雪浪>白雪塔>十八号>金玉良缘>如花似玉;叶柄中总酚含量的顺序:似荷莲>萍实艳>丹皂镏金>如花似玉>金星雪浪>金玉良缘>白雪塔>洛阳红>十八号;外植体材料的最佳水冲洗时间是1 h。
An experiment was conducted to study the total phenol content and the activation of Polyphenol oxidase(PPO) of nine tree peony eulvitars. This study focused on the relationship between the total phenol content, the activation of PPO and tissue browning. However, tissue browning is the key to affect the success of the tissue culture of tree peony. Explants propagated from nine culvitars were used. The total phenol content and the activity of Polyphenol oxidase in explants were determined. The results showed that the total phenol content of petiole was lower than that of leaf in nine culvitars. The order, which was total phenol content in leaves of nine tree peony cultivars, was cv. Si He Lian〉cv. Ping Shi Yan〉cv. Luo Yang Hong〉cv. Dan Zao Liu Jin〉cv. Jin Xing Xue Lang〉cv. Bai Xue Ta〉cv. Shi Ba Hao〉cv. Jin Yu Liang Yuan〉cv. Ru Hua Si Yu. The order of total phenol content in petiole was cv. Si He Lian〉cv. Ping Shi Yank〉 cv. Dan Zao Liu Jin〉cv. Ru Hua Si Yu〉cv. Jin Xing Xue Langley. Jin Yu Liang Yuan〉cv. Bai Xue Ta 〉cv. Luo Yang Hong〉cv. Shi Ba Hao. The total phenol content and the activation of PPO were reduced after lh water washing. They were lower than natural pretreatment. The best time of water washing was 1 h.
