摘要
目的观察藏药柳茶对单纯性肥胖大鼠脂质代谢的影响,初步探讨其作用机制。方法:复制单纯性肥胖大鼠模型,藏药柳茶水提取物灌胃给药8周后,检测体重、体长,计算Lee’s指数,分离并称取肾周及睾周脂肪重量;生化法检测血糖(GLU)、血总胆固醇(CHOL)、甘油三酯(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)的含量;放射免疫法检测血清瘦素(LEP)和胰岛素(INS);琼脂糖凝胶电泳检测脂肪细胞凋亡状况。结果:柳茶治疗后动物血糖、血脂含量明显降低,血瘦素和胰岛素水平下降。但实验未见到明显促脂肪细胞凋亡的作用。结论:藏药柳茶可明显抑制大鼠体重增长,其作用机制可能与改善瘦素和胰岛素抵抗、加快脂肪代谢有关。
Objective To study effects of the extract from Sibiraea angustata (Rchd.)Hand.-Mazz on the lipid metabolism in rats with simple obesity and to initially explore its mechanism. Methods Rat models of simple obesity were established and were given gastric gavage of the extract from Sibiraea angustata (Rchd.) Hand.-Mazz. After treat- ment for 8 weeks, the body weight and body length were measured for the calculation of the Lee's index. Then we separated and counted the fat around the kidney and testicle. Biochemical method was utilized to detect the contents of serum glucose (GLU),cholesterol (CHOL), triglyceride (TG), high-density lipoprotein (HDL), low-density lipoprotein (LDL), alanine aminotransferase (ALT)and aspartate aminotransferase (AST). Radioimmunoassay was used to detect leptin(LEP)and insulin(INS). Agarose gel electrophoresis was applied to detect the apotosis of the fat ceils. Results After treatment with the extract from Sibiraea angustata (Rchd.)Hand.-Mazz, the contents of GLU, blood lipid, ALT and AST in the rats decreased. So did the levels of LEP and INS . However, the effect on promoting fat cell apoptosis was not obvious. Conclusion The extract of Sibiraea angustata (Rchd.)Hand.-Mazz can inhibit the increase of rats body weight, and protect hepatocytes from damage, and its mechanism is probably related to relieving LEP and INS resistance, protecting hepatocytes and speeding lipid metabolism.
