摘要
目的研究传统药对远志-石菖蒲配伍后石菖蒲对远志兔体内药代动力学的影响。方法远志组和远志-石菖蒲(1∶1)配伍组分别灌胃给药,反相高效液相色谱法测定3,4,5-三甲氧基肉桂酸(TMCA)的血药浓度,DAS 2.0软件对结果进行统计分析。结果远志组和配伍组TMCA均符合二室开放模型,配伍组TMCA的t1/2Ka和t1/2α缩短,Cmax和Tmax增大,AUC(0-∞)增加,t1/2β增大,CL/F降低。结论石菖蒲使远志代谢产物TMCA在兔体内吸收加快,吸收量增大,达峰时间延迟,消除减慢。研究成果可为传统药对体内协同作用机制的研究提供思路。
Aim To study the effect of Acorus tatarinowii Schott on Pharmacokinetics of Polygala tenuifolia Willd.in the drug pairs of Polygala tenuifolia Willd and Acorus tatarinowii Schott.Methods P.tenuifolia and the drug pairs were separately orally administrated to health rabbit.The blood concentration of the 3,4,5-trimethoxycinnamylic acid was measured by HPLC method.The data were statistically analyzed by DAS 2.0.Results The pharmacokinetic parameters of TMCA conformed to the two-compartment open model in P.tenuifolia group and the drug pairs group.T1/2Ka,t1/2αand CL/F decreased,t1/2β prolonged,while Cmax,Tmax and AUC(0-∞) increased,after P.tenuifolia and A.tatarinowii were coadministrated.Conclusion A.tatarinowii improves the absorption of TMCA,slows down the elimination,increases the bioavailability.This research can provide reference for synergistic effect mechanism of drug pairs in vivo.
