摘要
目的探讨肝纤维化组织学量化诊断方法。方法用二甲基亚硝氨(DMN)诱导SD大鼠肝纤维化模型,常规肝组织切片,Mason三色和苦味酸天狼红染色,苦味酸固绿-天狼红染色比色测定,图像定量处理和半定量计分分析及羟脯氨酸(Hyp)含量测定。结果切片胶原比色测定、图像定量处理和半定量计分分析均与肝纤维化程度变化相一致,与肝组织Hyp含量具有良好的相关性,r分别为094、089和088,P均<001。结论切片胶原测定。
Objective To discuss the histological quantitative method of hepatic fibrosis. Methods Dimethylnitrosamine (DMN) was used to produce an SD rat model for hepatic fibrosis and the effects with histological technique and liver hydroxyproline (Hyp) content observed. Histology technique included HE stain, Masson stain, Picric acid-Sirus red stain, and Picric acid-Sirus red stain colorimetric technique. Then picture quantitative analysis technique and semi-quantitative scoring method was used to analyse the effect of colorimetric determination. Results There was better consistency between the change of hepatic fibrosis and the colorimetric determination of collagen content, picture quantitative determination, semi-quantitative scoring method. There was better correlationship between Hyp content and determining effects of these three methods. The coefficient 'r' of the three effects were 0 94, 0 89 and 0 88, respectively(P<0 01). Conclusion These results suggest that it is possible to use collagen content determination in slice, picture quantitative determination and semi-quantitative scoring for the quantitative diagnosis indices in hepatic fibrosis.
