摘要
目的探讨LPS等丝裂原所致THP-1细胞增殖分化的优化因素。方法利用MTT法及改良WST-8法测定THP-1细胞的增殖情况,并对THP-1细胞生长特性及其曲线进行测定,同时测定成熟THP-1细胞的吞噬功能。由此探讨LPS、PHA、ConA对PMA诱导成熟的THP-1细胞活化增殖的最佳浓度及作用时间曲线。结果WST-8法测定THP-1细胞增殖活性优于MTT法,比较敏感;THP-1细胞增殖测定表明LPS比PHA、ConA效果好;LPS浓度在1μg/ml、作用24h可充分诱导活化巨噬细胞,同时巨噬细胞具有吞噬鸡红细胞的能力。结论LPS可使巨噬细胞活化增殖,并且吞噬功能明显增强。
Objective To evaluate the optimization factors of LPS and other mitogens-induced THP-1 cell proliferation. Methods The WST-8 assay,a modification of MTT assay,was used to determine the THP-1 cell proliferation in experimental conditions and the phagocytosis of mature THP-1 cells. The optimal concentration and time of LPS, PHA and ConA inducing the activation of mature THP- 1 cells were investigated, and the proliferation curve was determined. The optimal concentration and time of LPS inducing macrophage were observed. Results WST-8 was better and more sensitive than the MTr method to determine the activation of THP-1 cell proli- feration. The effect of LPS inducing proliferation was better than that of PHA, ConA. The LPS fully induced the activation of macrophage at the concentration of 1 μg/ml for 24 h,and the activated giant macrophage had the phagocytosis for chicken red blood ceils. Conclusion LPS could activate the proliferation of macrophages and increase the phagocytosis.
