红曲霉菌胞外多糖提取及抗氧化活性测定 被引量:5

摘要 目的:红曲霉菌胞外多糖发酵提取优化,测定红曲霉菌胞外多糖清除DPPH能力,分析红曲胞外多糖糖链组成。方法:取摇床转速150r/min和120r/min,收集96h,102h,108h,120h的除细胞发酵液,以1/4、1/2、2/3、1/1,浓缩比减压蒸馏去细胞发酵液,95%的酒精分级醇沉,收集15%、30%、50%、75%醇沉沉淀物,苯酚-硫酸法测多糖含量并计算多糖得率;测定各胞外多糖的DPPH清除能力。结果:摇床转速150r/min多糖得率较高,96h发酵的多糖得率明显高于其它发酵时间,1/4浓缩,红曲霉胞外多糖(Exopolysaccharide,Eps)分布最均匀,2/3浓缩提取胞外多糖量最多;75%醇沉多糖清除DPPH最强。结论:不同的浓缩比和发酵时间对红曲霉菌胞外多糖得率有明显的影响;红曲霉菌胞外多糖有清除DPPH能力,以75%醇沉多糖作用为主。 Objective:To study the best conditions of Monascus EPS fermented and distilled,the effect of Monascus EPS Clearing DPPH and the main role of part of the,Analysis of the composition of its sugar chain.Methods:Check shaker speed 150r/min and 120r/min;Collected 96h,102h,108h,120h of the cell-free broth,then mixed concentrated vacuum distillation the cell-free broth with the proportion of 1∶4,1∶2,2∶3,1∶1,separate out polysaccharide with 95% of the alcohol,Collected 15%,30%,50%,75% alcohol precipitate,Polysaccharide content tested with phenol-sulfuric acid and Polysaccharide yield calculationed,Determination of EPS cleared effect of DPPH and TLC analysised Monascus EPS. Results:Shaker speed 150r/min got More Polysaccharides; 96h fermentation of polysaccharides yield was significantly higher than yield of other fermentation time; concentrate with the properation of 1∶4,Distribution of Eps was most well-distributed,concentrated and extracted with the properation of 2∶3,xould get large number of Eps; It cleared the m concentrate ost DPPH of the 75% alcohol precipitation polysaccharide.Monascus EPS,only a small amount of sugar chain of glucose.Conclusion:Shaker speed,Fermentation time and Concentrated broth impacted Monascus EPS yield; Monascus EPS could clear DPPH,75% alcohol precipitation polysaccharide is the main force of eliminate.
