不同掺锶浓度羟基磷灰石涂层对成骨细胞生物学行为的影响 被引量:9

摘要 目的:观察不同锶浓度掺锶羟基磷灰石涂层对成骨细胞附着、增殖和碱性磷酸酶(ALP)表达的影响。方法:采用微弧氧化方法制备掺锶原子数百分比[Sr/(Sr+Ca)]分别为2.22%,7.24%,15.15%和23.78%的涂层,将原代培养的成骨细胞接种到涂层表面,采用MTT方法检测30、60、90min时成骨细胞在材料表面的附着和2、3、7d时细胞在材料表面的增殖情况;并对第3天和第7天成骨细胞ALP活性进行检测。结果:成骨细胞在材料表面培养60min和90min时掺锶浓度为15.15%和23.78%组的细胞附着明显高于2.22%和7.24%组(P<0.05)。成骨细胞在材料表面培养24h和72h后15.15%和23.78%组的细胞增殖明显高于2.22%和7.24%组(P<0.05)。ALP活性检测显示7d时锶浓度为2.22%和7.24%组细胞的ALP活性高于其他组(P<0.05)。结论:随Sr含量的增高,涂层对成骨细胞的附着和增殖有明显的促进作用,掺锶浓度低的涂层体现了较好的ALP活性。 AIM:To evaluate the response of osteoblast to strontium-sincorperated hydroxyapatite(Sr-HA) coatings prepared by micro-arc oxidation(MAO) with different Sr concentrations.METHODS:MAO method was used to prepare coatings with different concentrations(2.22 %,7.24 %,15.15 % and 23.78 %) of strontium.Primary osteoblasts were plated directly on the coated surface.Cell adhesion was examined 30,60 and 90 min after cell plating.The cell proliferation was examined 1,3,7 d after plating by MTT method.Alkaline phosphatase(ALP) activity was assessed with ALP assay kit.RESULTS:15.15 % and 23.78 % Sr-HA enhanced osteoblasts adhesion and proliferation,while 7.24 % Sr-HA increased the values of ALP activity after 7 d culture.CNCLUSION:Sr-HA coatings containing relatively high concentration of strontium promote osteoblasts attachment and proliferation,while coatings with relatively low concentration of strontium have stronger ALP increasing activity.
