摘要
目的研究纤维蛋白原(Fg)Bβ-249C/T和Bcl-1G/A基因多态性与血浆Fg浓度、分子聚合活性等指标和脑梗死(CI)的关系。方法选取160例急性CI患者、114例陈旧CI患者和160例正常对照者,应用PCR方法进行Bβ-249C/T和Bcl-1G/A位点的基因多态性分析;采用微机辅助血浆Fg功能自动监测系统测定血浆Fg浓度、Fg单体聚合反应速率(FMPV)、最大光密度(Amax)、FMPV/Amax等。结果急性CI组Bβ-249C/T的CT+TT型FMPV/Amax低于CC型,Bcl-1G/A的GA+AA型Fg浓度、FMPV均高于GG型,陈旧CI组Bcl-1G/A的GA+AA型FMPV/Amax高于GG型,正常组Bcl-1G/A的GA+AA型Fg浓度高于GG型(P均<0.05)。结论FgBβBcl-1G/A变异基因型可调控血浆Fg含量,吸烟、血糖升高等因素可通过Bcl-1G/A变异基因型使血浆Fg浓度和FMPV的表达增强,Bcl-1G/A变异基因型是CI发病或复发的高危人群。
Objective To explore the relation of beta-fibrinogen gene -249C/T, Bcl-1G/A polymorphisms and plasma fibrinogen concentration, molecular polymerized function and cerebral infarction. Methods 160 acute cerebral infarction patients, 114 remote cerebral infarction patients and 160 normal control were to be chosen. Gene polymorphisms of Bβ -249C/T 和 Bcl-1G/A were analyzed by polymerase chain reaction-restriction fragment length polymorphism. The parameters illustrated Fg molecular reactivity, such as Fg concentration, fibrin monomer polymerize velocity( FMPV), absorbance maximum (Amax) , FMPV/Amax, were examined by microcomputer assistanted blood Fg function automatic monitoring system. Results In the acute cerebral infarction group, FMPV/Amax of CT + TT genotype of Bβ - 249C/T was lower than that of CC genotype, and Fg concentration, FMPV of GA + AA genotype of Bcl-IG/A was higher than that of GG genotype. In the group of remote cerebral infarction, FMPV/Amax of GA + AA genotype of Bcl-1G/A was higher than that of GG genotype. In the group of normal control, Fg concentration of GA + AA genotype of Bcl-1G/A was higher than that of GG genotype ( all P 〈 0.05 ). Conclusions Bcl-1 polymorphisms contributes to control Fg concentration. With the impact of smoking and hypergly-caemia, expression of Fg concentration and FMPV can be enhanced by variation genotype of Bcl-1. Someone who had variation genotype of Bcl-1 is high-risk people to trouble acute and recurring cerebral infarction.
