小菜蛾颗粒体病毒Px26基因功能的初步分析

摘要 用Px26和Px26-egfp融合基因瞬时表达质粒对多种昆虫传代细胞系进行转染实验,探讨Px26的功能属性.实验结果表明,Px26编码产物主要定位于被转染细胞的细胞膜上,在低pH诱导下未观察到被转染细胞膜融合现象. Or f26 (Px26) of Plutella xylostella granulovirus(PlxyGV) is a homolog of Agrotis segeturn granulovirus (AgseGV) f gene, which was recently shown being able to functionally substitute gp64 of Autographa California multiple nucleopolyhedrovirus (AcMNPV). Transient expression of Px26, fused with a copy of the gene of enhanced green fluorescent protein (EGFP), was perform in Sf9 and other four insect cell lines. It was shown that the Px26 EGFP mainly positioned on the cell membrane of the cells transfected with the transient expression plasmid of the fused gene Px26-egfp, but cell membrane fusion was not observed.
