摘要
目的:纯化细菌的精氨酸-tRNA合成酶。方法:培养大肠杆菌、酵母菌、嗜热脂肪芽胞杆菌,分别采用超声波破菌和丙酮破菌、层析、测活力峰。结果:酵母菌、大肠杆菌用不同和破菌方法纯化结果完全相同,丙酮破菌法显著提高嗜热脂肪芽胞杆菌ArgRS的纯化。结论:丙酮在破菌过程中去掉了嗜热脂肪芽胞杆菌体内的大量脂类化合物,提高了纯化效率。
Objective:Purification of bacterial Argining tRNA synthetase(ArgRS).Methods:The purification of ArgRS for Escherichia Coli.Yeast and Bacillus stearothermophilus(Bst) includ breaking bacterium in Ultrasonic and acetone.Chromtography and assaing activity.Results:Breaking Bst in acetone Heighten the purification efficiency of Bst's ArgRS.Conclusion:Because acetone can remove a lot of lipid chemical compound of Bst,this method can shorten the time and simplify the process of enzyme purification
