摘要
分别以偶氮方法和碳化二亚胺(EDC)方法将β2受体激动剂克伦特罗(CL)和塞曼特罗(CIM)连接到载体蛋白上,并以连接物免疫家兔,分别制备兔抗CL和CIM抗血清,经间接ELISA和间接抑制ELISA检测抗血清效价和特异性,并在此基础上建立了CL和CIM快速免疫检测方法。2种抗血清工作浓度分别为1∶10000和1∶5000。CL和CIM免疫检测范围分别为1.9ng~15.6μg和0.477ng~1.85μg。2种β2受体激动剂快速免疫检测方法的建立为监督畜牧生产上滥用CL和CIM,检测肉品CL和CIM残留提供了一种简便、灵敏、特异的手段。
Two β 2 agonists, Clenbuerol (CL) and Cimaterol (CIM) were conjugated with carrier proteins by diazotization and EDC respectively. These conjugates were used to immunize rabbits to prepare anti CL and anti CIM sera. The titers and the specificities of the sera were detected by indirect ELISA and inhibitory indirect ELISA. Then the sera were used to build up immunoassay methods for the two kinds of β 2 agonists. The work dilutions of two kinds of sera were 1∶10 000 and 1∶5000 respectively and the sensitivites of detection ranged from 1.9 ng to 15.6 μg and from 0.477 ng to 1.85 μg respectively. These immunoassay methods could be used to examine the overdue usage of CL and CIM in animal productions and to detect the residue of CL and CIM in meat products.
