摘要
利用疏基(-SH)能与金形成稳定的Au-S键,先将硫脲固定在金电极上,再利用交联剂乙二醛将乙肝抗体(HBsAb)固定在电极上,从而形成单分子膜,制成用于检测乙型肝炎表面抗原(HBsAg)的电流型免疫传感器.优化反应条件使反应时间缩短、稳定性好、灵敏度提高.通过循环伏安法考查电极的电化学特性,结果表明,该传感器的检测范围为5ug/L^200ug/L,检测下限为1.0 ug/L,可用于检测乙型肝炎表面抗原.
A novel electrochemical immunesensor for the detection of Hepatitis B surface antigen had been developed by means of self-assembly to immobilize Hepatitis B surface antibody on a gold electrode. At first, the modification of Au electrode with thiourea by chemical adsorption was achieved. Secondly, glyoxal was chemically adsorbed, finally the Hepatitis B surface antibody was immobilized on the Au electrode. A state of short reaction time, remarkable stability, and high sensitivity was gained when the reaction condition was optimized. The cyclic vohammetry was used to investigate the electrochemical reaction. The immunosensor responds rapidly to Hepatitis B surface antigen in the line range from 5 ug/L to 200ug/L, and a detection limit of 1 ug/L.
