茯苓水溶性多糖不同提取方法的比较研究 被引量:5

摘要 以茯苓水溶性多糖提取得率为考察指标,在分别通过单因素试验与正交试验优选茯苓水溶性多糖热水浸提、微波辅助提取以及超声辅助提取三种方法最优技术参数的基础上,比较研究了三种提取方法对同一茯苓原料水溶性多糖提取得率的影响。结果表明,热水浸提以水温100℃、时间5h、固液比1:25的工艺参数最佳,茯苓水溶性多糖提取得率为2.89%;微波辅助提取以中低火(231w)、时间60min、固液比1:25的工艺参数最佳,茯苓水溶性多糖提取得率为3.66%;超声辅助提取以40kHz强度下超声处理20rain、100℃热水浸提3h、固液比1:25的工艺参数最佳,茯苓水溶性多糖提取得率为3.02%;三种方法相比,以微波辅助提取最好,其茯苓水溶性多糖提取得率分别为热水浸提、超声辅助提取的1.27倍、1.22倍。 With the yield rate of water-soluble pdysaccharide as the final indicatar, hot water extraction method, microwave-assisted extraction method and ultrasonic-assisted extraction method were investigated and optimized by using single factor experiments and orthogonal experiments, and these three methods were comparatively studied with same material. By analysis of range, the optimized condition of hot water extraction method:extracting 5 h at 100 ℃, solid-to-liquid ratio being 1 : 25, the extraction rate of polysaccharide was 2.89% ; the optimized condition of microwave-assisted extraction method: processing 60min, power being 231 W, solid-to-liquid ratio being 1: 25, the extraction rate was 3.66% ; the optimized condition of ultrasonic-assisted extraction method:processing 20 min for ultrasonic treatment, frequency being 40 kHz,solid-to-liquid ratio was 1:25 ,then,extracting 3 h at 100 ℃ in hot water,the extraction rate was 3.02%. Microwave-assisted extraction method was the best, comparing with the other two, for extraction yield rate of the optimized microwave-assisted extraction method for Poria cocos water-soluble pelysaccharide was 1.27 times of the optimized hot water extraction method and 1.22 times of the optimized ultrasonic-assisted extraction method.
