摘要
运用硫酸钡比浊法测定龙须菜多糖的硫酸基含量,并对测定方法进行了研究。采用硫酸钡比浊法,测得龙须菜多糖硫酸基含量7.13%±0.09%。研究显示,反应试剂明胶和氯化钡-明胶溶液的不同处理方式对标准曲线有一定影响,同其他处理相比,溶液经离心处理后相同硫酸基含量吸光度值增大,12=0.999。稳定性试验表明,硫酸基在0—120μg线性关系良好(r=0.9994),同一样品在10~40min内检测值比较稳定。重复性试验显示,同一样品在25h内测定,差异较小(RSD=3.10%)。回收率为96.0%。
Barium sulfate turbidity was used for determining the sulfate content in the Gracilaria lemaneiformis polysaccarides(GCp) and the availability of this method was also studied. The sulfate content of GCp was detected to be 7.13% ± 0.09% by this method. The results showed that there were some influences on the stand curve by different prepara- tions of gelatin solution and BaCI2-gelatin solution. Compared with others, centrifugalizaion can increase the absorption (the same SO4^2_)and r^2 =0.999. The stability test indicated that there was a good linearity(r =0.9994)in the sulfate content range of 0-120 μg,and the results obtained from the same sample were stable within a time range of 10-40 min. The repetition test displayed that the detecting results of the same sample within 25 h had a little different(RSD =3.10% ). The recovery by this method was 96.0%.
