灰树花胞外多糖的分离纯化和性质分析

Purification and Physicochemical Characteristics Analysis of Exo-polysaccharides Produced by Grifola frondosa during Submerged Fermentation Process

摘要 灰树花经发酵培养后醇沉法得到粗多糖,对其进行脱蛋白、脱色和透析等初步纯化,并利用Sephadex G-200凝胶色谱进行二次纯化。采用光谱和色谱等方法对得到的单一多糖进行糖含量、旋光度、单糖组成等理化性质的测定。分离出的灰树花胞外多糖单组分GFP-1糖含量为80.9%,由核糖、木糖、甘露糖、葡萄糖和半乳糖组成,摩尔比为:0.06:0.32:1:0.06:0.11。红外光谱证实具有多个多糖的特征吸收峰。灰树花胞外多糖GFP-1是一种水溶性中性多糖。 This peper studied the isolation and purification of polysaccharides from Grifola frondosa, extracted and exopolysaccharides by alcohol sedimentation and fractionated it by Sephadex G-200 gel chromatography. Physicochemical characteristics analysis was studied by spectrometry and chromatography. A fraction GFP-1 was obtained. Detection showed that the content of total polysaccharides was 80.9%. GC analysis indicated that GFP-1 was composed of Rib, Xyl, Man, Glu and Gal with molar ratio of 0.06: 0.32:1: 0.06:0.11. FrIR showed that GFP-1 had several characteristic absorption peak of polysaccharides. GFP-1 was a kind of neutral water-soluble polysaecharide.
