摘要
目的:探讨葫芦素B抗血吸虫肝纤维化的作用及其机制。方法:方法60只小鼠随机分为3组。小鼠感染尾蚴6周后,以葫芦素B0.2mg/kg.d治疗8周,第14周末处死小鼠,留取肝脏组织,观察各组小鼠肝组织病理改变,测定其肝脏丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性以及血管内皮生长因子(VEGF)、VEGFmRNA、Ⅰ、Ⅲ型胶原的表达变化。结果:实验对照组小鼠出现明显肝纤维化,肝组织中SOD活性降低而MDA含量以及VEGFmRNA、VEGF、Ⅰ、Ⅲ型胶原表达均明显高于正常组。葫芦素B治疗可减轻肝纤维化程度,显著升高肝组织中SOD活性、降低MDA含量以及VEGFmRNA、VEGF、Ⅰ、Ⅲ型胶原表达水平,但仍未恢复正常。结论:葫芦素B可通过抑制肝纤维化组织VEGF表达,降低肝组织氧化应激水平而发挥抗血吸虫肝纤维化作用。
Objective：To study the inhibitory effect and mechanisms of cucurbitacin B on the liver fibrosis due to Schistosoma japonicum infection.Methods：Sixty mice were divided into three groups.Group A,group B were infected with Schistosoma japonicum.Group C were fed normally.After six weeks,group A was treated with cucurbitacin B 0.2mg/kg·d for for 8 weeks.Group B was taken as experimental control without any treatment.At 14th week,all mice were sacrificed and a part of liver tissues were preserved.The changes of hepatic histopathology were observed.The contents of MDA and the activity of SOD in liver as well as the expressions of hepatic VEGFmRNA,VEGF,typeⅠ and type Ⅲ collagen in mice infected with S.japonicum before and after treatments were measured.Results：There were evident hepatic fibrosis,being depressed the activity of SOD and heightened the expression of hepatic MDA,VEGFmRNA,VEGF,typeⅠ and type Ⅲ collagen in the experimental control.The therapy of cucurbitacin B could relieve the degree of hepatic fibrosis,reduce the contents of hepatic MDA,VEGFmRNA,VEGF,typeⅠ and type Ⅲ collagen obviously and greatly heighten the activity of SOD,but those did not get back to normal level.Conclusion：The inhibitory effect of cucurbitacin B on the liver fibrosis due to Schistosoma japonicum infection may be based on inhibiting the expressing of hepatic VEGF that lead to reducing the level of oxidative stress.
