脑源性神经营养因子在兔透镜诱导性近视眼视网膜神经节细胞中的表达

Expression of brain derived neurotrophic factor in retinal ganglion cells in myopia induced by lens of rabbits

摘要 目的观察发育期兔配戴凹透镜后眼球屈光度及眼轴长度的变化,检测视网膜神经节细胞中脑源性神经营养因子(brain derived neurotrophic factor,BDNF)阳性细胞的表达情况。方法健康大耳白兔20只,兔龄14d,随机分为A、B2组,左眼均配戴-10.00D凹透镜镜片为诱导眼,右眼为对照眼;A组凹透镜诱导后第60天时处死动物取材,B组在透镜诱导后第45天时去掉凹透镜继续饲养15d时处死动物取材。实验前及诱导后第15天、第30天、第45天、第60天分别测定双眼屈光度及眼轴长度,处死动物后摘除眼球。第60天同时检测兔视网膜神经节细胞BDNF的阳性细胞表达情况。结果诱导后第60天时A组诱导出(-1.84±0.46)D的近视,诱导眼较对照眼眼轴延长(0.96±0.52)mm;B组诱导出(-1.13±0.35)D的近视,B组在取下透镜镜片后近视进展明显减缓;但是在去掉透镜镜片15d后,诱导眼屈光度与对照眼(1.34±0.44)D相比差异仍具有显著统计学意义(P<0.01)。A组诱导后第60天时BD-NF阳性细胞数诱导眼(5.25±1.03)个较对照眼(10.63±1.68)个明显减少,差异具有显著统计学意义(P<0.01);B组BDNF阳性细胞数诱导眼(6.75±1.49)个与对照眼(10.38±1.77)个相比明显减少,差异也有显著统计学意义(P<0.01)。结论视网膜神经节细胞BDNF表达可能参与了离焦性近视的形成。 Objective To observe the changes of ocular refraction and axial length on developmental rabbits after wearing concave lens,and detect the positive expression of brain derived neurotrophic factor(BDNF) in retinal ganglion cells.Methods Twenty healthy white rabbits with 14 days old were randomly divided into two groups,the left eyes wearing-10.00 D concaves lens were set as induced eyes,and the right as control eyes.The cases in group A were sacrificed and drawn the materials after induction on the 60th day,and group B after removed concave lens to keep induction for 15 days after induction on the 45th day.The diopter and axial length in dual eyes were detected before experiment and on the 15th day,30th day,45th day and 60th day after induction respectively,the cases were sacrificed and the eyeballs were enucleated,the positive expression of BDNF in retinal ganglion cells was examined on the 60th day after induction.Results After induction on the 60th day,the myopia in group A and group B were (-1.84±0.46)D and (-1.13±0.35)D respectively,the axial length of induced eyes was prolonged (0.96±0.52)mm compared with that of control eyes in group A;In group B the progression of myopia was obvious slow after removed lens,but at 15 days after that,the refraction in control eyes was (1.34±0.44)D,there was sig-nificant difference in refraction between induced eyes and control eyes(P〈0.01).After induction on the 60th day,the positive cells of BDNF in induced eyes in group A,which was (5.25±1.03)cells,obviously decreased compared with that in control eyes,which was (10.63±1.68)cells,there was significant difference(P〈0.01),but that in induced eyes in group B,which was (6.75±1.49)cells,obviously decreased compared with that in control eyes,which was (10.38±1.77)cells,there was also significant difference(P〈0.01).Conclusion The expression of BNDF in retinal ganglion cells layer may take part in the formation of lens induced myopia.
