Optimization of AFLP-PCR Reaction System in Coconut

摘要 以椰子叶片为试材,通过对AFLP反应体系中的几个关键因素进行优化,建立了适宜椰子的AFLP分子标记反应体系。本研究最终确定了20μL PCR反应体系的最佳条件为:预扩增产物稀释20倍液2μL,dNTPs 0.5μL,引物1.0μL,Taq聚合酶0.2μL,MgCl22.0μL。该优化体系稳定可靠,适合应用于椰子AFLP分析。 Total genomic DNA is extracted from the leaflets of coconut, and the suitable AFLP - PCR reaction system of coconut is established. The optimum reaction system (20 uL ) consists of 2 uL 20 times dilution of the pre - amplification product, 0.5 uL dNTPs, 1.0uL primer pairs, 0.2uL Taq DNA polymerse, 2.0 uL MgCl2. The result shows that this system is stable and reliable, and can be used in the AFLP analysis of coconut.
