不同培养条件对油茶愈伤组织和芽诱导的影响 被引量:14

摘要 [目的]为油茶的快速繁殖提供技术参考。[方法]以油茶品种岑软2号和岑软3号的鲜嫩带芽的茎段和顶芽为外植体,分别设置不同消毒时间,9组培养基激素浓度配置,2个光照条件,进行组织培养。[结果]在其他条件一致的情况下,不同的外植体消毒方式、培养基激素配置和光照条件对油茶愈伤组织和芽的诱导的影响各异。采用0.1%HgCl2对外植体消毒6min的诱导效果最好,岑软2号和岑软3号的成活率分别达71%和73%;激素配置为6-BA2.0mg/L和NAA0.2mg/L时,愈伤组织和芽的诱导效果达到最佳,岑软2号和岑软3号的诱导率分别为47.8%、48.5%;接种后直接进行光照培养的效果最好,岑软2号和岑软3号的愈伤组织和芽的诱导率分别为45%、62%。[结论]最佳条件:0.1%HgCl2消毒6min;激素配置为6-BA2.0mg/L和NAA0.2mg/L;接种后直接进行光照培养。 [Objective]The study aimed to provide the technical reference to rapid propagation of Camellia oleifera.[Method]With the stem and terminal bud of cenruan 2 and cenruan 3 as explant,respectively set up different disinfection time,9 groups hormone concentration configuration,2 light condition,tissue culture was conducted.[Result]Under the same condition,different disinfection method,hormone concentration configuration,illumination condition had the different induction effect on callus and buds of Camellia oleifera.It was found that with 0.1% HgCl2 for 6 minutes had the best effect,survival rate of cenruan 2 and cenruan 3 were 71% and 73%,respectively;6-BA 2.0 mg/L and NAA 0.2 mg/L had the best effect,induction rate of cenruan 2 and cenruan 3 were 47.8%,48.5%,respectively;lighting directly after inoculation had the best effect,induction rate of cenruan 2 and cenruan 3 were 45%,62%,respectively.[Conclusion]The best conditions was:disinfection with 0.1% HgCl2 for 6 min;6-BA 2.0 mg/L and NAA 0.2 mg/L;lighting directly after inoculation.
