摘要
目的:研究鸡骨骼肌肌钙蛋白T同分异构体的表达。方法:对2d龄鸡进行自体肌肉移植,将胸肌植入腿肌、腿肌植入胸肌、前背阔肌植入胸肌和腿肌后,用光镜和电镜观察组织形态,用双向电泳和免疫标记检测肌钙蛋白T同分异构体的变化。结果:发现供体肌纤维在移植后短期内退化,然后逐渐增生形成新的肌纤维,新生肌纤维细胞与受体组织肌细胞有不完全融合现象;新生肌具有表达原组织肌钙蛋白T同分异构体的特性,这种特性一直维持至观察期的第6个月。
Objective: To study the expression of troponin T isoforms in chicken skeletal muscle. Methods: The change of histology and the expression of troponin T isoforms of autoplastic transplanted muscles were examined, respectively, by light microscope and electron microscope, two dimensional SDS polyacrylamide gel electrophoresis and immunoblotting with antitroponin T antibodies after transplantation of breast muscle into leg muscle, leg muscle into breast muscle, and anterior latissimus dorsi into breast and leg muscle. Results: The transplanted muscle produced degeneration during short period after transplantation and then began multiplication and grew up neonatal muscle fibers. The regenerated muscles expressed the specifical troponin T isoforms to the donor tissue on the 178th day after the transplantation. Furthermore, the phenomenon of a noncomplete fusion between the muscle fibers of the donor tissue and the host tissue was observed. Conclusion: The expression speciality of the troponin T isoforms in one type of chicken skeletal muscles is primarily fixed in the cell lineage.
