摘要
运用CO2超临界萃取装置同时对荷叶、山楂、决明子和泽泻的复方物进行萃取,用Box-Bebnken响应曲面设计法,以萃取压力、萃取温度、夹带剂用量为自变量,以总生物碱、总蒽醌、总三萜类物质得率3个指标用多指标实验全概率法处理后所得的全概率公式分为响应指标,用回归方程的方差分析检验模型的准确性。最优化的条件为萃取压力30.93MPa,萃取温度50.04℃,夹带剂用量2.16mL/g。在此条件下提取3次结果与理论预测值相近。因此,基于多指标实验全概率公式分的响应曲面法所得的优化提取工艺参数准确可靠,具有实用价值,可用于荷叶复方降脂产品的开发。
Supercritical carbon dioxide extraction was employed to extract alkaloids, anthraquinones and total triterpenes simultaneously from lotus leaf compound prescription(LCP) which composed of lotus leaves, hawthorn fruit, cassia seeds and Alisma orientale.The optimal conditions for extraction were determined by response surface methodology.Box-Behnken design was applied to evaluate the effects of three independent variables (pressure, temperature and co-solvent amount)on the above active substances yields of LCP, with the response values carried out by the multiple guidelines grading method.Correlation analysis of the mathematical-regression model indicated that a quadratic polynomial model could be employed to optimize the supercritical carbon dioxide extraction of LCP.The optimal conditions obtained were pressure 30.93MPa,temperature 50.04℃, and co-solvent amount 2.16mL/g. Under these optimal conditions, the experimental values agreed with the predicted values, indicating a high goodness of fit of the model used.
