摘要
[目的]探讨DNMT1和UHRF1在胃癌的形成和转化中的表达特点和可能的生物学作用。[方法]提取CIMP型H(High)的胃癌细胞系HGC-27和CIMP型为L(low)的胃癌细胞系MGC803,BGC823及AGS的核酸样品进行UHRF1和DNMT1表达的分析。[结果]DNMT1在MGC823和HGC-27表达水平明显高于其他胃癌细胞系(P<0.01);UHRF1在HGC-27中有表达水平高于其他3个胃癌细胞系(P<0.01)。[结论]胃癌细胞DNA的甲基化不但和DNMT1有关而且和UHRF1也有着密切关系。
[ Objective] To elucidate DNMT1 and UHRF1 expression patterns of gastric cancers and their potential roles in gastrocarcinogenesis. [ Methods] Samples of RNAs were isolated from established human gastric cancer cell lines such as MGC803, BGC823 and AGS with CIMP- low and HGC -27 with CIMP- high DNA methylation patterns, and UHRF1 and DNMT1 expression was analyzed by RT -PCR. [ Results] DNMT1 was up -regulated in MGC823 and HGC cells, while it was expressed in low level in MGC803 and HGC - 27 ( P 〈 0.01 ). UHRF1 expression level in CIMP - H HGC - 27 cells was higher than that in the other three cell lines (CIMP- L),P 〈0.01. [ Conclusion] DNA methylation in gastric cancer cells is correlated either with DNMT1 or with UHRF1 expression.
